Prolia(denosumab) andZometa(zoledronic acid) are used to treatboneloss (osteoporosis)inwomen who are at high risk for bonefractureaftermenopause.

Zometais also used to increase bone mass in men with osteoporosis. Zometa is also used to treat Paget'sdisease, highbloodlevels ofcalciumcaused bycancer(hypercalcemiaofmalignancy),multiple myeloma(a type ofbone marrowcancer), or metastaticbone cancer.

Prolia and Zometa belong to different drug classes. Prolia is amonoclonal antibodyand Zometa is abisphosphonate.

Commonside effectsof Prolia include:

• low calcium levels (especially if you havekidneyproblems),
• weakness,
• constipation,
• back pain,
• musclepain,
• pain in yourarmsand legs,
• anemia,
• diarrhea, or
• skinproblems (eczema, blisters,dry skin、脱皮、红肿,itching, smallbumps).

You may also be more likely to get a seriousinfection, such as a skin,ear,stomach/gut, orbladder infectionwhile taking Prolia. Tell yourdoctorif you develop signs of infection, such as:

• fever/chills,night sweats,
• red/swollen/tender/warm skin (with or withoutpus),
• severe stomach orabdominal pain,
• ear pain or drainage, trouble hearing,
• frequent/painful/burning urination, or
• pink/bloodyurine.
• severe itching, burning, rask, blistering, peeling, or dryness of the skin,
• cough,
• shortness of breath,
• pinpoint purple or red spots under your skin,
• flusymptoms, or
• weight loss.

Tell your doctor if your experience serious side effects of Prolia includingjawpain, new or unusualthigh/hip/groinpain, or bone/joint/muscle pain.

Prolia may interact with steroids or cancer medicine,cyclosporine,sirolimus, tacrolimus, basiliximab,muromonab-CD3,mycophenolatemofetil,azathioprine,leflunomide, or etanercept.

Zometamay interact withdiuretics(waterpills),lithium,methotrexate, pain orarthritismedicines, medicines used to treatulcerative colitis, medicines used to preventorgantransplantrejection, IV antibiotics,antiviralmedicines, or cancer medicines. Tell your doctor all medications and supplements you are taking.

Prolia should be administered by a doctor. The recommended dose of Prolia is 60mgadministered as a singlesubcutaneous(under the skin) injection once every 6 months.

Zometais administered underphysiciansupervision. The maximum recommended dose of Zometa in hypercalcemia of malignancy or in patients with multiple myeloma and metastatic bone lesions from solid tumors is 4 mg as a single-doseintravenousinfusion over no less than 15 minutes. Duration of treatment varies depending on theconditionbeing treated.
